چکیده

GSK-3 (Glycogen synthase kinase-3) is a serine/threonine protein kinase involved in various physiological processes and its inhibitors have been evaluated as promising drug candidates for severe pathologies. In this paper a PLS regression analysis was used, to explore the relationship between field of descriptors (as independent variable) and biological activities (pIC50 as dependent variables). The main purpose of this work is to develop a robust QSAR model for predicting inhibitory activity of the 109 indirubin derivatives and better understanding of structural features of these compounds and their relation with the inhibitory activity using the Dragon descriptors. The statistical qualities of the final model have been assessed by several parameters such as: the cumulative sum of squares of correlation coefficient RY(CUM)=0.872, the cumulative fraction of the total variation of the Y values that can be predicted for all the extracted principal components by cross validation Q2(CUM) = 0.804,R2 supplied by the Golbraikh-Tropsha external validation method and Yrandomization test.

GSK-3-selective inhibitors derived from Tyrian purple indirubins.

Gastropod mollusks have been used for over 2500 years to produce the "Tyrian purple" dye made famous by the Phoenicians. This dye is constituted of mixed bromine-substituted indigo and indirubin isomers. Among these, the new natural product 6-bromoindirubin and its synthetic, cell-permeable derivative, 6-bromoindirubin-3'-oxime (BIO), display remarkable selective inhibition of glycogen synthase...

Molecular dynamics study of the inhibitory effects of ChEMBL474807 on the enzymes GSK-3β and CDK-2

Indirubin derivatives and analogs comprise a significant group of ATP-competitive inhibitors. The inhibitory effects of ChEMBL474807 (1-(4-amino-1,2,5-oxadiazol-3-yl)-5-(piperidin-1-ylmethyl)-N'-(pyridin-4-ylmethylene)-1H-1,2,3-triazole-4-carbohydrazide) on two enzymes, namely glycogen synthase kinase-3β (GSK-3β) and cyclin-dependent kinase-2 (CDK-2), were analyzed. The close resemblance of the...

Efficacy of small-molecule glycogen synthase kinase-3 inhibitors in the postnatal rat model of tau hyperphosphorylation.

BACKGROUND AND PURPOSE Glycogen synthase kinase-3 (GSK-3) affects neuropathological events associated with Alzheimeŕs disease (AD) such as hyperphosphorylation of the protein, tau. GSK-3beta expression, enzyme activity and tau phosphorylated at AD-relevant epitopes are elevated in juvenile rodent brains. Here, we assess five GSK-3beta inhibitors and lithium in lowering phosphorylated tau (p-tau...

Inhibition of glycogen synthase kinase-3beta protects dopaminergic neurons from MPTP toxicity.

Glycogen synthase kinase-3beta (GSK-3beta) is closely involved in neuronal apoptosis and pathogenesis of many neurodegenerative diseases, such as Alzheimer's disease. However, whether GSK-3beta mediates apoptosis of dopaminergic neurons in Parkinson's disease remains elusive. In this study, using 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced Parkinsonism models, we investigated wh...

Glycogen synthase kinase-3β may contribute to neuroprotective effects of Sargassum oligocystum against amyloid-beta in neuronal SH-SY5Y cells

Glycogen synthase kinase (GSK)-3β mediates amyloid-beta (Aβ) and oxidative stress-induced neurotoxicity in neurodegenerative disorders. Natural products with antioxidant activity, such as Sargassum (S.) oligocystum may modulate GSK-3β enzyme and protect against Aβ-induced neurotoxicity. Therefore, we aimed to assess the neuroprotective effects of a methanolic extract of S. oligocystum against A...
